neuroleptics disposal from dopamine receptors inside brain tissue after stopping

hi im lookin for answer to this question , after stopping neuroleptic it takes 5times the half life to be disposed from the body but is it the same for brain tissue ?

i heard that once neueroleptic enters brain tissue and start there job blocking dopamine its stays inside the brain forever or takes realy long time to be disposed many years

so im confused do you have a link to study research about this matter ?

will neuroleptic disposed by the half life rule for a drug from the brain or there are different mechanism of disposal in the brain than the whole body and it takes much longer time

your answer will be highly appreciated

The 5 half life rule applies in any body tissue, but the rate of diffusion (or transport) into or out of that tissue may affect clearance. The brain is a special case because it has a layer of tissue specifically designed to keep xenobiotics (read: drugs, poisons, etc) out. But even then, clearance from the brain should be reflected in blood levels.

This is bullshit. Some of the side effects are permanent, but the drug itself is cleared quickly when you discontinue it. Ask anyone who's ever stopped taking their antipsychotics and relapsed three months later.

You're confusing the effects of a medication with its presence.

I believe that the drug isn't removed in the length of the half-life, because the one I'm on (Ziprasidone) has a 7-hour half-life and I take it every 12. Confusing, isn't it?

Yeah, but Ziprasidone reaches max serum concentration only after 6-8 hours, so it will give 12 hours coverage.

I was jonesing for more every six hours. Withdrawal effects. Chills, goose bumps. It was a horrible drug for me. I continued to cycle and got psychotic even though I was on other stabilizers. Awful. Bad, bad medicine for me. Two trials, the second one worse. Fortunately, it is the only AAP to be like that for me, too.

The half life of a drug is not 50% of the time it takes to clear it completely. It's the point at which 50% of the drug has been cleared. The difference is subtle, but important:

Examine the blue line on this plot. Pretend it represents the plasma concentration of a drug after a single dose, and 1 is the peak concentration. The half life of this drug will be the point where the concentration reaches .5, which happens to be .7 on the X (time) axis. But the drug will take another 4.3... uh... units, before its concentration approaches zero. So, there's a very long period of time after the half-life where the drug is still present in measurable quantities. Half of the time to completely clear a drug is not a very useful value to a doctor - the drug's effect is usually gone long before then.

The function in the graph above is exponential. No drug follows a perfect exponential decay model in its metabolism, but most will track one pretty well.

Half life is not necessarily related to duration of action. Many drugs persist in the body long after their effects dissipate, and others have effects that persist long after the drug is gone.
