Does your Welby wear off at night?

Recommended Posts

Anyone on Wellbutrin XL have problems at night? seems like the stuff wears off around 6pm. I've always had trouble with night time depression though so its hard for me to say. Just curious. My days are normal, nights just get me in negative thought patterns.

yes, i do feel like it wears off in the early evening.  i've been considering split dosing, last time i was on WB i had to do that.  also when i wake up in the morning i feel a distinct absence of the drug - it's like i'm not even taking anything until it kicks in mid-morning.  i question my sanity in the morning until it starts working, then i remember i'm going to be okay again for the next eight hours or so.

I'm on wellbutrin xr and have been for years, and never noticed much of a difference in the evenings. But I've now started a job that requires me to wake up quite early and I've noticed it wears off in the evening. I might try taking half my dose in the morning and the other half a few hours later or something.

I am no longer on wellbutrin SR, as I wrote above.  I am on the IR.  I take it twice a day and I find it doesn't affect sleep ... my sleep is messed up, but it has been messed up since 2006.  For me, wellbutrin keeps me awake during the day and a little more focused.  Without it I would be dragging with my eyes half shut.  (Provigil also helps with this).

The clincher is that the Wellbutrin XL generics don't really do a good job of evenly distributing the drug. At least in my experience with two different ones. It wasn't until I got on Aplenzin that I actually felt like it was lasting 24 hours. And I knew it was because I would wake up ready to go and it didn't take me long to really get going. Aplenzin has also made me a lot less jittery.

I get my data from the PI which states a steady state is achieved  after 8 days of continuous dosing.

...You clearly do not understand what steady state is. Here, let me explain.

https://en.wikipedia.org/wiki/Pharmacokinetics

In pharmacokinetics, steady state refers to the situation where the overall intake of a drug is fairly in dynamic equilibrium with its elimination.

In other words, this means serum levels at exactly identical time points during the day remain approximately the same irregardless of the number of days separating the exactly identical time points during the day. You will notice in the graphs that I posted, the serum concentrations regress to a relatively steady and predictable level towards the end of each day. These graphs are at steady state and are thus essentially an infinite loop assuming drug intake quantities/times are kept exactly the same. The consumption and elimination are in balance, so no major fluctuations in serum levels would be expected to occur assuming drug intake quantities/times are kept exactly the same.

I hope this helps explain the concept of steady state to you.
